A Simple Method of Preparative Electrophoresis.
نویسندگان
چکیده
A method of preparative electrophoresis is described. A volume of approximately 1 ml. of protein solution can be applied with the cell used. A “marker” solution, applied adjacent to the sample to be separated, indicates the positions of the protein bands during the migration. The procedure has the advantages of a high degree of recovery, simplicity of operation, and utilization of an electrophoretic cell which can also be applied to other electrophoretic procedures.

Preparative SDS-PAGE Electroelution for Rapid Purification of Alkyl Hydroperoxide Reductase from Helicobacter pylori
BACKGROUND Alkyl hydroperoxide reductase (AhpC) of Helicobacter pylori is considered as a diagnostic antigen. Therefore, this antigen can be used to detect H. pylori infection by stool immunoassays such as ELISA. The aim of this study was to simplify the AhpC protein purification procedures. METHODS For whole cell protein extraction, the bacterial cells were ruptured by octly-β-D glucopyranos...
متن کاملGram-preparative protein fractionation by isotachophoresis: isolation of human growth hormone isohormones.
Steady-state stacking (isotachophoresis) provides a preparative method in zone electrophoresis for the isolation of proteins in gram amounts. Up to four components were separated in 1000-fold larger quantities than those used in conventional electrophoresis. Separation occurs in order of free electrophoretic mobility. Recovery is nearly quantitative. The method was shown to be capable of resolv...
متن کاملSeparation and isolation of fusion protein using a new native preparative PAGE device.
A human serum albumin and Thymosin α1 (HSA-Tα1) fusion protein was designed and over-expressed in Pichia pastoris. To purify the fusion protein, a new native preparative electrophoresis system that involved a modified device with a sample receiving chamber, and an assay method with Coomassie Blue G-250 tracing the collection of the protein of interest. In this device, two gels were run in paral...
متن کاملA Method for Applying Haemolysates on Starch Blocks for Electrophoresis.
Electrophoresis of haemolysates on a starch block, which was first described by Kunkel and Wallenius (1955), provides an excellent method for separating and quantitating the various normal and abnormal haemoglobins. Since relatively large amounts of haemoglobin solutions can be applied it is a useful preparative method for isolating a particular haemoglobin for further analysis. One of the impo...
متن کاملGradient gel electrophoresis of human plasma lipoproteins.
A rapid and simple method for separating and identifying plasma lipoproteins is described. The technique utilizes the “molecular sieving” properties of continuous concave gradient electrophoresis on a slab of polyacrylamide (4 to 26%) gel. Gradient gel electrophoresis is particularly suitable for separating lipoproteins according to their molecular weight. The technique can be used to demonstra...
